Ascorbate caused a dose-dependent increase in sister-chromatid exchanges (SCEs) in Chinese hamster ovary (CHO) cells and in human lymphocytes. Moreover, in the DNA synthesis inhibition test with HeLa cells, ascorbate gave results typical of DNA-damaging chemicals. Catalase reduced SCE induction by ascorbate, prevented its cytotoxicity in CHO cells, and prevented its effect on HeLa DNA synthesis. Ascorbate reduced induction of SCE in CHO cells by N-methylN′-nitrosoguanidine (MNNG) by direct inactivation of MNNG.  相似文献

Summary The effect of salinity on the quality of various agricultural crops has not yet been explored much. This information is very important to Israel due to the increasing use of saline water for irrigation. This paper reports the effect of saline irrigation water on the quality, especially the taste, of several crops. Fruits from a processing tomato cultivar exposed to various degrees of salinity had higher values for total soluble solids (TSS) and acidity than their controls. The yield of fruit after saline water irrigation is lower, but this is offset by the nigher fruit quality and its consequent higher value. Melon fruits from plants subjected to saline water scored higher in taste than their controls when the fruits were analyzed fresh. After 3–4 weeks of storage at room temperature, there was no longer any difference in taste. Even though salinity slightly increased the TSS content, this did not correlate with the taste scores. Iceberg lettuce grown with saline water did not significantly differ in taste from its control, even when the sensitive triangle taste test was used. The same was true for peanuts. Thus, for these two crops no advantage of better quality would compensate for possible lower yields. Salinity had little effect on the yield of two varieties of Chinese cabbage, but increased the frequency of tipburn.  相似文献

Acrodysostosis is an extremely rare disorder at birth, that is, characterized by skeletal dysplasia with short stature and midfacial hypoplasia, which has been reported to be caused by PDE4D and PRKAR1A gene mutations. Here, a Chinese boy with acrodysostosis, ventricular septal defect, and pulmonary hypertension was recruited for our study, and his clinical and biochemical characteristics were analyzed. A novel de novo heterozygous missense mutation (NM_001104631: c.2030A>C, p.Tyr677Ser) of the PDE4D gene was detected by whole exome sequencing and confirmed by Sanger sequencing. The c.2030A>C (p.Tyr677Ser) variant was located in exon 15 of the PDE4D gene, predicted to be damaging by a functional prediction program and shown to be highly conserved among many species. Further functional analysis showed that the p.Tyr677Ser substitution changes the function of the PDE4D protein, affects its subcellular localization in transfected cells, increases PDE4 activity in the regulation of cAMP signaling and affects cell proliferation. Our study identified a novel de novo PDE4D mutation in acrodysostosis of Chinese origin that not only contributes a deeper appreciation of the phenotypic characteristics of patients with PDE4D mutations but also expands the spectrum of PDE4D mutations.  相似文献

通过对秦岭山区中国大鲵(Andrias davidianus)栖息地生境因子调查、统计,利用R语言分析了各因子与大鲵生境选择的相关性,得出研究结果:秦岭山区影响大鲵生存的主要因子为栖息地类型(相关系数r=0.98),其次是水温(相关系数r=-0.8)、河岸坡度(r=-0.6)和p H (r=-0.6);浊度(相关系数r=0.5)、电导率(r=0.49)、DO(r=0.4)、人为干扰(r=0.35)和海拔(r=0.31)对大鲵分布影响不大。研究结果为探讨中国大鲵对野生环境的适应性选择提供了参考。  相似文献

Prolonged endoplasmic reticulum (ER) stress reduces protein synthesis and induces apoptosis in mammalian cells. When dimethyl sulfoxide (DMSO), a specific monoclonal antibody productivity (qmAb)‐enhancing reagent, is added to recombinant Chinese hamster ovary (rCHO) cell cultures (GSR cell line), it induces ER stress and apoptosis in a dose‐dependent manner. To determine an effective ER stress inhibitor, three ER stress inhibitors (BiP inducer X [BIX], tauroursodeoxycholic acid, and carbazole) are examined and BIX shows the best production performance. Coaddition of BIX (50 μm ) with DMSO extends the culture longevity and enhances qmAb. As a result, the maximum mAb concentration is significantly increased with improved galactosylation. Coaddition of BIX significantly increases the expression level of binding immunoglobulin protein (BiP) followed by increased expression of chaperones (calnexin and GRP94) and galactosyltransferase. Furthermore, the expression levels of CHOP, a well‐known ER stress marker, and cleaved caspase‐3 are significantly reduced, suggesting that BIX addition reduces ER stress‐induced cell death by relieving ER stress. The beneficial effect of BIX on mAb production is also demonstrated with another qmAb‐enhancing reagent (sodium butyrate) and a different rCHO cell line (CS13‐1.00). Taken together, BIX is an effective ER stress inhibitor that can be used to increase mAb production in rCHO cells.  相似文献

The increasingdemandfor biopharmaceutical products drives the search for efficient cell factories that are able to sustainably support rapid growth, high productivity, and product quality. As these depend on energy generation, here the genomic variation in nuclear genes associated with mitochondria and energy metabolism and the mitochondrial genome of 14 cell lines is investigated. The variants called enable reliable tracing of lineages. Unique sequence variations are observed in cell lines adapted to grow in protein‐free media, enriched in signaling pathways or mitogen‐activated protein kinase 3. High‐producing cell lines bear unique mutations in nicotinamide adenine dinucleotide (NADH) dehydrogenase (ND2 and ND4) and in peroxisomal acyl‐CoA synthetase (ACSL4), involved in lipid metabolism. As phenotypes are determined not only by functional mutations, but also by the exquisite regulation of expression patterns, it is not surprising that ≈50% of the genes investigated here are found to be differentially methylated and thus epigenetically controlled, enabling a clear distinction of high producers, and cells adapted to a minimal, glutamine (Gln)‐free medium. Similar pathways are enriched as those identified by genome variation. This strengthens the hypothesis that these phenomena act together to define cell behavior.  相似文献

ObjectiveTo study the changes of ginsenoside content in different proportion of Panax ginseng-Angelica sinensis (GA) co-decoction, and to explore the amelioration of hematopoietic function in mice after combined use of the two drugs. The active ingredient profiles in P. ginseng single decoction and co-decoction of GA were determined by high performance liquid chromatography (HPLC). The experimental pharmacology method was used to explore the effect of GA co-decoction on the hematopoietic function of chemotherapy mice.ResultsThe active ingredient profiles of the co-decoction of GA significantly changed compared with those of the single decoction. Compared with GA1:0 (single decoction of Panax ginseng), the routine ginsenosides of all proportions of GA decreased significantly, but the proportion of rare ginsenosides increased significantly. The changes of contents of rare ginsenosides of GA were basically consistent with the trends of effects on the myelosuppression induced by CY. Compared with the model group, GA significantly increased the number of bone marrow nucleated cells, thymus index, peripheral blood leukocytes and platelets, and significantly reduced the spleen index. Moreover, GA could promote G1 phase bone marrow cells to enter the cell cycle, increase the proportion of S phase cells and G2/M phase cells, and increase the cell proliferation index.ConclusionGA can ameliorate the hematopoietic function of mice after chemotherapy, and GA2:3, GA3:2 were the best, which may be due to the changes of the pharmacodynamic material basis of GA after compatibility. All these results implied that GA may be an ideal drug and food supplement for the treatment of toxic and side effects of chemotherapeutic drugs.  相似文献

2000-2015年中国陆地生态系统蒸散时空变化及其影响因素   总被引:1,自引:0,他引:1  
准确量化区域蒸散时空格局及其影响因素对理解陆地生态系统碳水循环具有十分重要的意义。近年来中国经历了严重的空气污染及气候波动,亟须探讨蒸散的时空变化及其影响因素。基于PT-JPL(Priestly-Taylor Jet Propulsion Laboratory)模型,集成遥感数据和气象数据模拟了中国陆地生态系统2000-2015年蒸散,并分析其时空变化及影响因素。结果表明:1)参数优化后PT-JPL模型可解释蒸散年际变化的68%,优于原始模型及MODIS蒸散产品;2)中国地区多年平均蒸散为440.16 mm/a,呈东南沿海到西北内陆逐渐递减的空间格局;3)2000-2015年蒸散整体呈轻微下降趋势(slope=6.48 Gt/a,P=0.17),但具有年代际差异,2000-2010年中国地区蒸散呈显著下降趋势(slope=21.05,P < 0.01),占全国蒸散总量45.05%的内蒙古地区、甘新地区、黄土高原地区及青藏地区解释了61.88%的年际变化;2010-2015年呈轻微上升趋势(slope=10.48,P=0.71),各地区均无显著变化趋势;4)辐射主导了蒸散的年代际差异,分别解释了2010年前后蒸散下降及上升趋势的51.45%、85.26%。蒸散呈显著变化趋势的内蒙古地区、黄土高原地区及青藏地区主要受辐射控制,甘新地区主要受降水和温度的影响。  相似文献
